บทคัดย่อ: โลมาอิรวดี หรือโลมาหัวบาตร (Orcalella brevirostris) เป็นสัตว์ที่รับความคุ้มครองในระดับชาติ และนานาชาติ ดังนั้นจึงจำเป็นต้องมีเครื่องมือที่ถูกต้องแม่นยำในการติดตามตัวสัตว์ในสถานแสดงโลมา สถานเพาะพันธุ์ และในธรรมชาติเพื่อเตรียมพร้อมสำหรับการอนุรักษ์ในอนาคต การศึกษานี้จึงทดสอบประสิทธิภาพของลายพิมพ์ดีเอ็นเอไมโครแซทเทลไลท์ 11 ตำแหน่ง (8 ตำแหน่งพัฒนามาจากโลมาปากขวด และ 3 ตำแหน่งพัฒนาขึ้นมาใหม่) ในการบ่งชี้อัตลักษณ์ของโลมาอิรวดีจากอ่าวไทย โดยใช้ตัวอย่างซากเกยตื้นจากอ่าวไทย (n=36) และทะเลอันดามัน (n=3) ที่รวบรวมระหว่างปี พ.ศ. 2537-2553 ผลการศึกษา พบว่าเครื่องหมายที่พัฒนาขึ้นมาใหม่ 3 ตำแหน่ง มีระดับความหลากหลายในระดับปานกลาง (จำนวนอัลลิลต่อตำแหน่ง=8-18) และลายพิมพ์ดีเอ็นเอที่ประกอบไปด้วยเครื่องหมายทั้ง 11 ตำแหน่ง สามารถบ่งชี้อัตลักษณ์ของโลมาอิรวดีจากอ่าวไทยได้ โดยมีค่าความน่าจะเป็นที่ลายพิมพ์ดีเอ็นเอของสัตว์สองตัวจะซ้ำกัน (PI) เท่ากับ 2 x 10-12 และ 3.3x10-5 สำหรับกรณีที่สัตว์ไม่ได้มีความสัมพันธ์ทางเครือญาติ และสัตว์ที่มีความสัมพันธ์ ตามลำดับ นอกจากนี้ยังพบว่า ลายพิมพ์อีเอ็นเอที่ประกอบไปด้วยเครื่องหมายอย่างน้อย 6 ตำแหน่งก็เพียงพอที่จะ บ่งชี้อัตลักษณ์ของโลมาอิรวดีจากอ่าวไทยได้ (ค่า Plsibs เท่ากับ 2.0x10-3 หรือ 2 ตัวใน 1,000 ตัว) ระดับความหลากหลายทางพันธุกรรมของกลุ่มตัวอย่างส่วนใหญ่ (อ่าวไทยตอนกลาง, Gulf-C; ตราด, TRT; อ่าวไทยตอนในฝั่งตะวันออกและฝั่งตะวันตก, UG-E, UG-W) อยู่ในระดับใกล้เคียงกันระหว่างกลุ่มโลมาอิรวดี (ค่า allelic richness เฉลี่ยทุกตำแหน่ง= 3.18-3.92 และ ค่า observed heterozygosity = 0.58-0.69) ส่วนกลุ่มตัวอย่างจากอันดามัน มีค่า allelic richness เฉลี่ยทุกตำแหน่งน้อยที่สุดคือ 2.73 เมื่อพิจารณาความแตกต่างทางพันธุกรรมระหว่างกลุ่มพบว่า ตัวอย่างจากอันดามัน มีแนวโน้มที่จะต่างกับตัวอย่างจากอ่าวไทย และพบความแตกต่างระหว่างกลุ่มตัวอย่างภายในอ่าวไทย โดยตัวอย่าง Gulf-C และ TRT ต่างกับตัวอย่างจากอ่าวไทยตอนใน (UG-E, UG-W) ส่วนความต่างทางพันธุกรรมระหว่าง Gulf-C และ TRT และระหว่าง UG-E และ UG-W ยังไม่สามารถหาข้อสรุปได้อย่างแน่ชัดเนื่องจากหลายวิธีการทดสอบยังให้คำตอบที่ไม่ตรงกัน การจัดกลุ่มตัวอย่างใหม่ตามระดับความคล้ายกันของจีโนไทป์ของสัตว์แต่ละตัว โดย Discriminant analysis of principal components สามารถแบ่งกลุ่มได้ 2-3 กลุ่ม ที่ประกอบด้วย (1) ตัวอย่างส่วนใหญ่อ่าวตราด+อ่าวไทยตอนกลาง (2) ตัวอย่างส่วนใหญ่จากอ่าวไทยตอนในทั้งสองฝั่ง และ (3) ตละจำนวนจากตราด ชลบุรี และจันทบุรี ซึ่งอาจบ่งชี้แหล่งพันธุกรรมอื่นที่แตกต่างจาก 2 กลุ่มข้างต้น Irrawaddy dolphin (Orcaella brevirostris) is a protected species under national and international laws. To prepare for future conversation measures, we need a standardized tool to identify individuals in captivity and in the wild. This study evaluated the effectiveness of DNA fingerprinting profiles consisting of 11 microsatellite loci for individual identification. Eight loci were newly developed for Irrawaddy dolphin under the present study. I analyzed multilocus genotypes of 39 stranded individuals collected during 2003 to 2011 from the Gulf of Thailand (n=36) and Andaman Sea (n=3). The novel markers revealed a reasonable level of polymorphisms (allele per locus =8-18). A combination of eleven loci were adequate for identification of individuals obtained from the Gulf of Thailand, with a probability of identiy (P) value of 2 x 10-12 and 3.3x10-5 for unrelated and sibs (Plsibs) respectively. A DNA fingerprint that can differentiate individuals from the Gulf of Thailand would require at least six loci (Plsibs = 2.0x10-3 ). Most samples (four samples from Gulf of Thailand, Gulf-C, TRT, UG-E and UG-W) had moderate levels of genetic diversity (allelic richness averaged across loci=3.18-3.92; observed heterozygosity averaged across loci = 0.58-0.69). The Andaman sample had the lowest value of alletic richness (2.73). I detected population genetic structure among samples. The Andaman sample was likely to be different from the Gulf of Thailand samples. Samples with Gulf of Thailand were also genetically distinct with Gulf-C & TRT (central Gulf of Thailand and Trat bay) being different from UG-E & UG-W (inner Gulf of Thailand). The difference between Gulf-C and TRT as well as between UG-E and UG-W were inconclusive as different tests offered slightly different insigts. An indivudal assignment approach, Discriminant Analysis of Principal component (DAPC), offered an interesting insight. All individuals can be grouped into two to three clusters, (1) most individuals from TRT and Gulf-C, (2) most individuals from UG-E and UG-W and (3) a mixture of few individuals from UG-E and TRT. The third group may represent another genetic source as it is close proximity to anther coastal population of Cambodia.
