However, excess NO and PGE2 production has been reported to invole in a variety of inflammation-related diseases. In our on continuous effort to discover a novel anti-inflammatory agent, 1,1-bis-[2-(5-methylfuryl)]-4-nitrophenyl methane (JJSD-9), a bis(heteroaryl) alkane, has been shown its potential in anti-infammatory activity. The present study aimed to inveatigate the mechanism underlying its anti-inflammatory activity in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. JJSD-9 exhibited the potent inhibitory effect on NO and PGE2 production with IC50 values of 42.4+-1.3 and 36.1+- 6.3 um, respectively. JJSD-9 down-regulated the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) mRNA and protein in a concentration-dependent manner but it did not affect the activity of iNOS and COX-2 enzyme. Moreover, the compound did not show in vitroNO redical scavenging activity. JJSD-9 did not suppress nuclear translocation of NF-KB p65 subunit and NF-KB DNA binding activity. Likewise, the compound significantly reduced the phosphorylation of p38 MAPK but not ERK in LPS-induced cells. Taken together, these results suggest that JJSD-9 exhibited anti-inflammatory effect in RAW 264.7 macrophages mediated partially by decrease inphosphorylation of p38 MAPK leading to the reduction of iNOS and COX-2 expression, consequently, decrease in NO and PGE2 production. Thus, the JJSD-9 would be a lead compound for the development of novel anti-inflammatory agents.
