ทำการตรวจสอบความหลากหลายทางสายพันธุ์ของปูทะเลสกุล Scylla จากจังหวัดชลบุรี และจังหวัดตราด ในพื้นที่ภาคตะวันออกของประเทศไทยโดยการศึกษาความแตกต่างทางสัณฐานวิทยา (Morphology differences) การวิเคราะห์ความแตกต่างของสัดส่วนรยางค์ (Morphometric analysis) และ การศึกษาลักษณะความแตกต่างทางพันธุกรรมโดยเทคนิค Random amplified polymorphic DNA (RAPD) พบว่า จากการศึกษาลักษณะความแตกต่างทางสัณฐานวิทยาในปูทะเลสกุล Scylla จำนวน 336 ตัวอย่าง ที่มีขนาดความกว้างของกระดองขนาด 90-130 มิลลิเมตร สามารถแบ่งกลุ่มปูทะเล ออกเป็น 3 ชนิดได้แก่ ปูแดงหรือปูดำ (S. serrata) ปูขาว (S. oceanica) และปูเขียว (S. tranquebarica) พบว่าลักษณะที่มีความแตกต่างระหว่างชนิดมากกว่า 70% ของตัวอย่างทั้งหมด คือลักษณะสีที่ปรากฏบนส่วนขากรรไกร (Maxilliped), ลวดลายบนขาว่ายน้ำ, เส้นข้างกระดอง, หนามอันกลางบนข้อศอก, ลักษณะหนามบริเวณร่องกระดองตา และ ลักษณะสีลวดลายบนก้าม การวิเคราะห์ความแตกต่างของสัดส่วนรยางค์โดยวิธี Two ways ANOVA analysis พบว่าลักษณะของสัดส่วน AL/ICL และ ICW/ECW มีความแตกต่างกันอย่างมีนัยสำคัญ (p<0.05)
การศึกษาทางพันธุกรรมโดยเทคนิค RAPD ได้คัดเลือกไพรเมอร์ขนาด 10-15 นิวคลีโอไทด์ จำนวน 28 ไพรเมอร์และนำ 3 ไพรเมอร์ คือ UBC456, UBC457 และ YNZ22 ไปใช้ในการตรวจสอบความผันแปรของดีเอ็นเอ จากตังอย่างปูทะเลสกุล Scylla ทั้งสิ้น 85 ตัวอย่าง แบ่งเป็น ปูแดงในจังหวัด จันทบุรี 13 ตัวอย่าง จังหวัดตราด 20 ตัวอย่าง ปูขาว จันทบุรี 19 ตัวอย่าง จังหวัดตราด 11 ตัวอย่าง ปูเขียว จันทบุรี 4 ตัวอย่าง และจังหวัดตราด 18 ตัวอย่าง พบว่า มีแถบดีเอ็นเอที่เกิดจากไพรเมอร์ ทั้ง 3 ตัว จำนวน 92 แถบ มีขนาดอยู่ในช่วง 370-3000 คู่เบส ซึ่งมีความหลากหลายของแถบดีเอ็นเอเท่ากับ 70.37, 67.86, 77.59, 74.55, 47.92, และ 64.81% ตามลำดับ เมื่อนำค่าระยะห่างทางสายพันธุกรรม (genetic distance) มาสร้าง phylogenetic trees แสดงความสัมพันธ์ของปูแต่ละตัวอย่างประชากรพบว่าปูทะเลทั้ง 3 ชนิด มีลักษณะทางพันธุกรรมที่แตกต่างกันและไม่พบการแลกเปลี่ยนทางพันธุกรรมเกิดขึ้นระหว่างชนิดปูทะเล
Taxonomy and genetic diversity of mud crabs (genus Scylla) collected from Chantaburi and Trat Provinces in Eastern of Thailand were examined. The result from morphological analysis of 336 individuals (N = 132 for S. serrata, N = 157 for S. oceanica and N = 47 for S. tranquebarica) indicated significant differences in investigated characteristics between mud crab species. These included colour of maxilliped, pleopod and swimming line patterns, carapace lines, wrist spines and frontal teeth which clearly showed differences in more than 70% of all investigated mud crabs individuals. Base on mophometric analysis, result from two way ANOVA showed that the AL/ICL and ICW/ECW ratios of each mud crab species were statistically significant differences from each other (p<0.05)
Randomly amplified polymorphic DNA-Polymerase chain reaction (RAPD-PCR) was used for determination of genetic variability in S.serrata (N = 33), S. oceanica (N = 30) and S. tranquebarica (N = 22). A total number of28 primers (10-15 nucleotides) was screened. Of which three primers (UBC456, UBC457 and YNZ22) were highly informative. Ninety-two RAPD bands which molecular sizes between 370-3000 bp were observed. The percentage of polymorphic band was 47.925 – 77.595. A phylogenic tree base on genetic distances among pairs of individuals indicated three reproductively isolated group corresponding to S. serrata, S. oceanica and S. tranquebarica. Moreover, no shared RAPD genotypes were found indicating the apparent absence of genetic exchanges between mud crabs species in eastern Thailand should be regarded as three different species rather than different morphotypes. |
